What MOTS-c is and where it acts
MOTS-c is a 16-amino-acid peptide encoded in the mitochondrial 12S rRNA gene; its primary target organ appears to be skeletal muscle.[7]
In cells, it inhibits the folate cycle and its linked purine biosynthesis, which leads to AMPK activation.[7]
A review of its mechanism describes insulin sensitisation, enhanced glucose use and targeting of the folate-AICAR-AMPK pathway.[8]
What it did downstream (animals and cells)
Treated mice were protected from insulin resistance caused by ageing or a high-fat diet, and from diet-induced obesity.[7]
In non-obese diabetic mice, MOTS-c modulated T cell function by regulating TCR/mTORC1 signalling.[9]
In muscle cells and obese mice, MOTS-c lowered myostatin, acting through AKT phosphorylation and inhibition of the FOXO1 transcription factor.[10]
In immobilised mice, MOTS-c at 15 mg/kg/day limited muscle loss to about 5% against about 15% in untreated immobilised mice, an animal result.[11]
What has been measured in people
In humans, exercise induces endogenous MOTS-c expression in skeletal muscle and in circulation.[12]
In a human study, acute endurance exercise only showed a trend towards raising circulating MOTS-c.[13]
In people with type 1 diabetes, serum MOTS-c was lower than in healthy controls.[9]
In breast cancer patients, circulating MOTS-c did not change significantly with 24 weeks of chemotherapy with or without metformin.[8]

Common questions
How long does MOTS-c act in the body?
We found no published half-life for MOTS-c (PubMed search, 11 Oct 2026), so how long it acts is unknown.
How does MOTS-c work?
Through the folate cycle, purine synthesis and AMPK activation, shown in cells and mice.[7]
How long for MOTS-c to work?
No cited study reports a time to effect in people; the human papers measured the body's own MOTS-c, not treatment.[13]
