The three receptors retatrutide acts on

LY3437943, the development name of retatrutide, is described as a single peptide that is an agonist at the glucagon, GIP and GLP-1 receptors.[2]

In laboratory (in vitro) testing, LY3437943 showed balanced glucagon-receptor and GLP-1-receptor activity but more GIP-receptor activity.[2]

What the extra receptors seem to add

In obese mice, the glucagon-receptor part added increased energy expenditure on top of calorie-intake reduction driven by the other two receptors.[2]

In human liver cells in a dish, glucagon and retatrutide both reduced ANGPTL3/8 secretion, and a glucagon-receptor blocking antibody prevented that.[3]

The authors of that lipid study suggest glucagon-receptor agonism could lower ANGPTL3/8 and so contribute to the triglyceride and LDL-C falls.[3]

What was seen in people

In adults with type 2 diabetes, higher retatrutide doses lowered perceived hunger and disinhibition compared with placebo, and larger weight loss went with lower hunger.[4]

In a liver substudy, liver-fat falls were related to body weight, abdominal fat and markers of insulin sensitivity and lipid metabolism.[5]

Total fat mass in the diabetes substudy fell by 26.1% on the pooled 8 mg groups and by 23.2% on 12 mg, versus 4.5% on placebo.[6]

Why the injections are weekly

In the phase 1b trial the pharmacokinetics were dose proportional and the half-life was about 6 days.[1]

The first-in-human summary said the pharmacokinetic profile supported weekly dosing, with weight reduction persisting to day 43 after a single dose.[2]

What is not yet shown in people

The 2 Oct 2026 PubMed ledger for retatrutide with energy-expenditure and metabolic-rate terms lists 6 human-only papers, so a direct human measurement of how much of the effect comes from each receptor is not described in the cached abstracts.

Common questions

How long does Retatrutide act in the body?

The half-life values we found are approximately 6 days (subcutaneous). A half-life is how long the amount in the body takes to fall by half; it is not the same as how long an effect lasts.

How does retatrutide work?

Retatrutide is a single peptide that activates the glucagon, GIP and GLP-1 receptors, and the animal and cell work attributes extra energy expenditure and lipid effects to the glucagon receptor.[2]

What is the half-life of retatrutide?

The phase 1b trial reported a half-life of approximately 6 days.[1]
