Claim: it causes large weight loss
Adults with obesity but no diabetes lost a mean of 25.0% of body weight on the top dose in the phase 3 trial, against 3.9% on placebo.[2]
In the phase 2 obesity trial, the mean weight change at 48 weeks was -24.2% in the 12 mg group, compared with -2.1% on placebo.[1]
A network meta-analysis of weight-loss drugs found retatrutide carried the highest adverse-event risk among those compared.[17]
Claim: it lowers blood sugar
Adults with type 2 diabetes on diet and exercise alone saw HbA1c fall 1.94% on the top retatrutide dose against 0.81% on placebo in that monotherapy trial.[5]
The phase 2 diabetes trial said retatrutide reduced HbA1c more than 1.5 mg dulaglutide in its 8 mg slow-escalation and 12 mg groups.[11]
Claim: it eases knee pain and protects the heart
In the knee osteoarthritis subgroup of the phase 3 obesity trial, the largest pain-score fall on retatrutide was 3.6 points against 1.9 on placebo (hybrid estimand).[2]
For the heart, the evidence found is blood-pressure and lipid measurements pooled across trials, not heart events: systolic pressure fell by 6.79 mmHg on average.[18]
Claim: it clears fatty liver and calms inflammation
Liver fat fell by 82.4% on 12 mg and 81.4% on 8 mg in the 24-week fatty-liver substudy, while placebo moved by +0.3%.[10]
High-sensitivity C-reactive protein fell by 54.8% in the obesity study without diabetes, but the diabetes study did not show the same fall.[6]
Claim: it builds muscle and speeds metabolism
The body-composition substudy concluded that the share of lean mass lost within the weight lost was similar to other obesity treatments, which is lean-mass data and not muscle gain.[7]
Appetite, not metabolic rate, is what the diabetes questionnaire study measured: perceived hunger and the tendency to overeat were reduced at higher doses.[8]
