Evidence for cognition

Each row is an outcome related to cognition from a published study, strongest design first. Animal and cell results are labelled and are not human results. User reports are not counted as evidence here.

How strong is it?

Strongest evidence:Animal evidence, 2 of 4Journal of Huntington's disease 2024, PMID 38489193

A human randomized trial outweighs any number of animal studies. Animal and cell results show a mechanism worth testing, not that it works in people.

What memory means

MedlinePlus describes memory as the process of storing and then remembering information.[4]

Cognition outcomes measured in animals

The early dihexa paper reports that modified analogs kept the ability to reverse scopolamine-induced Morris water maze deficits and to augment hippocampal synaptogenesis, in animal models.[3]

In APP/PS1 mice the authors report that dihexa could be given orally and crossed the blood-brain barrier, and that AngIV tissue levels rose.[2]

The mouse study links the effect to the PI3K/AKT signalling pathway, which a PI3K inhibitor reversed.[2]

In the rat Huntington's-like model, 3-nitropropionic acid impaired spatial learning and memory consolidation, and the compound did not prevent it.[1]

What is missing

The early paper concludes only that dihexa 'may have therapeutic potential' for disorders such as Alzheimer's disease, based on animal data.[3]

Common questions

Does Dihexa help with cognition?

The strongest study we summarise for cognition is animal evidence, across 3 studies with an outcome for cognition. That is not proof it works in people.

Is Dihexa approved for cognition?

No. Dihexa is not FDA approved for any use, including cognition.

Does dihexa improve memory in people?

No human study is cited here; the memory results come from rats and mice, and one rat study found no protection against induced deficits.[1]
