Cagrilintide Results: What Studies Measured and When
What published studies measured for Cagrilintide, when those outcomes were measured, and what is still unproven.
What studies measured
Each outcome below comes from a published study, with its type and size, strongest design first. Animal and cell results are labelled as such and are not human results.
| Outcome | Result | Study type | Source |
|---|---|---|---|
| Pharmacokinetics in renal and hepatic impairment | Cagrilintide total exposure and peak concentration were similar across renal and hepatic function groups; the authors conclude dose adjustment is not warranted. | Human controlled trial | Clinical pharmacokinetics 2026, PMID 42228334 |
| QTc interval | No clinically relevant QTcF prolongation after the last 4.5 mg dose; upper 90% confidence limits of the placebo-adjusted change were below 10 ms. | Human RCTn = 105 | Diabetes, obesity & metabolism 2024, PMID 39279639 |
| Body weight at week 26 vs placebo | Weight reductions were 6·0%-10·8% across cagrilintide doses 0·3-4·5 mg versus 3·0% with placebo (trial product estimand). | Human RCT | Lancet 2021, PMID 34798060 |
| Cagrilintide 4.5 mg vs liraglutide 3.0 mg | Cagrilintide 4·5 mg gave a 10·8% weight reduction versus 9·0% with liraglutide 3·0 mg. | Human RCT | Lancet 2021, PMID 34798060 |
| Body weight at week 20 (with semaglutide 2.4 mg) | Combined with semaglutide, body weight fell by 15·7% (cagrilintide 1·2 mg) and 17·1% (2·4 mg) versus 9·8% with pooled placebo at week 20. | Human RCTn = 96 | Lancet 2021, PMID 33894838 |
| Body weight with cagrilintide monotherapy | Cagrilintide monotherapy reduced body weight by a mean difference of -6.08% (-5.89 kg) and also reduced blood pressure, without meaningful HbA1c improvement. | Meta-analysisn = 5425 | Annals of medicine and surgery 2026, PMID 42583410 |
| Body weight in diet-induced obese mice | Wild-type cagrilintide-treated mice lost -3.4 g of body weight. | Animal · mouse (not a human result) | EBioMedicine 2025, PMID 40609154 |
| Receptor dependence | Cagrilintide's weight-lowering effect depended on the amylin receptors AMY1R and AMY3R in mice. | Animal · mouse (not a human result) | EBioMedicine 2025, PMID 40609154 |
| Receptor pharmacology | AM833 (cagrilintide) showed a distinct pharmacological profile across receptor binding, activation and regulation measures compared with other agonists. | In vitro (not a human result) | The Journal of pharmacology and experimental therapeutics 2021, PMID 33727283 |
- Cagrilintide is an investigational amylin analogue (Novo Nordisk), not FDA-approved as of 2 October 2026. Phase 3 monotherapy trials RENEW 1 (NCT07220642) and RENEW 2 (NCT07220759) are registered as active, not recruiting; no results have been published in PubMed. The evidence here is a phase 2 monotherapy RCT plus mechanistic, pharmacokinetic and safety studies. Most phase 3 data are for the CagriSema combination (see the CagriSema page).
- No cagrilintide-specific evidence was found for blood pressure beyond a pooled meta-analysis, and no cagrilintide-specific data for diabetes, liver health or cholesterol were extracted.
- The phase 1b trial (PMID 33894838) combines cagrilintide with semaglutide 2.4 mg in every active arm, so its weight data are for the combination.
- The phase 2 trial (PMID 34798060) reports 706 participants randomised to cagrilintide plus 99 to liraglutide and 101 to placebo; no single total is given.
- Half-life is 159-195 h, from the combination phase 1b trial. No stability or storage study was found.
Typical timeline
When the studies above measured their outcomes. This is the study schedule, not a promise of when anyone will notice a change.
| When | What was measured | Study type | Source |
|---|---|---|---|
| 26 weeks | Primary endpoint: percentage change in body weight | Human RCT | Lancet 2021, PMID 34798060 |
| Week 19-20 | Pharmacokinetic sampling window (last dose to end of treatment) | Human RCTn = 96 | Lancet 2021, PMID 33894838 |

Common questions
How long does Cagrilintide take to work?
The studies we summarise measured outcomes at set points, for example Primary endpoint: percentage change in body weight at 26 weeks; Pharmacokinetic sampling window (last dose to end of treatment) at Week 19-20. Timelines outside studies vary.
Do you show Cagrilintide before and after photos?
No. Photos can't be verified, rarely show dose, diet or other drugs, and only the most dramatic ones get posted. We summarize measured outcomes instead.
