Orforglipron half-life
A half-life depends on the route (under the skin, into a vein, by mouth or nasal spray) and the species it was measured in, so we report each value with its route and source.
| Route | Half-life | Measured in | Source |
|---|---|---|---|
| oral | approximately 29 to 49 hours | People | FDA label (Foundayo) |
| oral | 48.1 to 67.5 hours (day 28, 2-24 mg, healthy adults) | People | Diabetes, obesity & metabolism 2023, PMID 37344954 |
| oral | 29 to 49 hours (week 12, type 2 diabetes) | People | Diabetes, obesity & metabolism 2023, PMID 37264711 |
| As labelled | approximately 29 to 49 hours (oral) | People | FDA label (Foundayo) |
What half-life means
Half-life is the time it takes for the amount of a drug in the blood to fall by half. After one half-life, about 50% is left; after two, about 25%; after four to five, only a few percent remain.
- A long half-life lets a drug be dosed less often, for example once a week.
- A short half-life means levels rise and fall quickly, which is why some peptides are given daily or more often.
- Effects can outlast blood levels, so half-life alone does not tell you how long a drug works.
What it means for dosing frequency
Dosing intervals used in published studies of Orforglipron:
- once-daily, oral (Human RCT; Lancet 2026, PMID 41275875)
- once per day, oral (Human controlled trial; Lancet 2026, PMID 41765029)
- once-daily, oral (Human controlled trial; Lancet 2026, PMID 42259339)
- once-daily, oral (Human RCT; Obesity pillars 2026, PMID 42577069)
Changing the interval changes how much drug builds up between doses, so it is not a free choice.
Drug tests and sport
Not on the WADA Prohibited ListGLP-1 and GIP agonists are not named anywhere on the 2026 list. This one is an approved medicine, so the catch-all S0 (which only covers drugs with no approval anywhere) does not apply to it.
Anti-doping labs use tests built for specific substances. Whether a given test detects Orforglipron, and for how long, is not reported in the anti-doping literature we checked (as of 2 Oct 2026).
Label half-life
For the labelled tablet, the document lists the time for drug levels to halve as roughly 29 to 49 hours after an oral dose.[3]
Half-life in trials
In healthy adults, mean half-life was 24.6 to 35.3 hours after a single 0.3-6 mg dose.[4]
On Day 28 of dosing, mean half-life was 48.1 to 67.5 hours across the 2-24 mg range.[4]
In adults with type 2 diabetes, mean half-life at Week 12 ranged from 29 to 49 hours.[5]
Food and interactions
The label reports no clinically relevant food effect on exposure.[3]
A food-effect paper found half-life and median time to peak comparable between fed and fasted states.[10]
The label says strong CYP3A4 inhibitors significantly raise orforglipron exposure while inducers reduce it.[3]
Common questions
How long does orforglipron stay in your system?
The label gives an elimination half-life of approximately 29 to 49 hours.[3]
What is the half-life of Orforglipron?
Approximately 29 to 49 hours (oral); 48.1 to 67.5 hours (day 28, 2-24 mg, healthy adults) (oral), from the sources cited below.
Is Orforglipron on the WADA prohibited list?
No, it is not listed. GLP-1 and GIP agonists are not named anywhere on the 2026 list. This one is an approved medicine, so the catch-all S0 (which only covers drugs with no approval anywhere) does not apply to it.
