Mechanism per the label

The Foundayo label says it is a GLP-1 receptor agonist that binds to and activates the human GLP-1 receptor.[1]

It describes GLP-1 as a physiological regulator of appetite and caloric intake.[1]

In animal studies, orforglipron reached and activated neurons in brain regions that regulate appetite and food intake.[1]

Not a peptide

The phase 2 obesity paper calls it a nonpeptide GLP-1 receptor agonist given once daily by mouth.[4]

Why once daily

The first-in-human paper concluded its long half-life of 25 to 68 hours allows once-daily oral dosing without water and food restrictions.[2]

The label says steady-state exposure is reached after approximately 1 week of once-daily use.[1]

Common questions

How long does Orforglipron act in the body?

The half-life values we found are approximately 29 to 49 hours (oral); 48.1 to 67.5 hours (day 28, 2-24 mg, healthy adults) (oral). A half-life is how long the amount in the body takes to fall by half; it is not the same as how long an effect lasts.

How does orforglipron work?

It activates the human GLP-1 receptor, which regulates appetite and food intake.[1]
