Claimed: weight loss from a daily pill
In a 3127-patient phase 3 trial in adults with obesity, mean body weight changed -7.5% with 6 mg, -8.4% with 12 mg and -11.2% with 36 mg of orforglipron against -2.1% with placebo at week 72.[6]
The Foundayo label approves it as an adjunct to diet and activity to reduce excess body weight and maintain weight reduction in adults with obesity or overweight with a weight-related condition.[18]
Claimed: better blood sugar
In a phase 2 trial in adults with type 2 diabetes, HbA1c changed by up to -2.10% with orforglipron against -0.43% with placebo and -1.10% with dulaglutide at week 26.[11]
Against dapagliflozin, HbA1c fell 1.23%, 1.50% and 1.56% with orforglipron 3 mg, 12 mg and 36 mg versus 0.81% with dapagliflozin 10 mg.[3]
Claimed: heart and blood-pressure benefits
In the obesity trial, waist circumference, systolic blood pressure, triglycerides and non-HDL cholesterol improved significantly with orforglipron compared with placebo.[6]
The label records the other side: a mean increase in heart rate of 4 to 5 beats per minute versus 0.5 with placebo.[18]
Claimed: lower inflammation
A pooled analysis of two phase 2 trials reported placebo-adjusted decreases in blood pressure, LDL cholesterol, triglycerides, ApoB, ApoC3 and hsCRP, an inflammation marker, with orforglipron.[8]
Honest limits
The head-to-head trial with oral semaglutide found more gastrointestinal events, discontinuations and pulse-rate increase with orforglipron.[2]
In a maintenance study, participants who reached a weight plateau kept a model-based estimate of 74.7% of their weight reduction on orforglipron.[4]

Common questions
Is Orforglipron proven to work?
The strongest study we summarise for Orforglipron is human rct evidence, among 15 studies. A trial result is not an approval; see the legal status.
Does orforglipron work?
Phase 3 trials reported larger weight and HbA1c changes than placebo, and the drug carries an FDA label for weight management.[6]
