Evidence for diabetes
Each row is an outcome related to diabetes from a published study, strongest design first. Animal and cell results are labelled and are not human results. User reports are not counted as evidence here.
| Outcome | Result | Study type | Source |
|---|---|---|---|
| Body weight change | In people with diabetes, bodyweight changed by -5·1% (6 mg), -7·0% (12 mg) and -9·6% (36 mg) with orforglipron vs -2·5% with placebo at week 72. | Human RCTn = 1613 | Lancet 2026, PMID 41275875 |

How strong is it?
A human randomized trial outweighs any number of animal studies. Animal and cell results show a mechanism worth testing, not that it works in people.
Diabetes in plain terms
MedlinePlus explains that insulin is a hormone made by the pancreas.[7]
Type 2 diabetes trials
In adults with early type 2 diabetes, orforglipron significantly reduced the glycated hemoglobin level over a period of 40 weeks.[4]
Against oral semaglutide at week 52, HbA1c fell 1.71% with orforglipron 12 mg and 1.91% with 36 mg, versus 1.23% with semaglutide 7 mg and 1.47% with 14 mg.[2]
Against dapagliflozin, every orforglipron dose was non-inferior in reducing HbA1c at week 40.[3]
In adults with type 2 diabetes, 1613 were randomly assigned after dose escalation to orforglipron 6 mg (n=329), 12 mg (n=332), 36 mg (n=322) or placebo (n=630).[1]
Registered HbA1c measures
The phase 2 record NCT05048719 registered change from baseline in HbA1c compared with placebo.[8]
The ATTAIN-2 record NCT05872620 registered change from baseline in hemoglobin A1c.[9]

Does Orforglipron help with diabetes?
The strongest study we summarise for diabetes is human rct evidence, across 6 studies with an outcome for diabetes. A trial result is not an approval; see the legal status.
Is Orforglipron approved for diabetes?
Orforglipron is FDA approved (Foundayo, Apr 2026). Which conditions that approval covers is set by the label; use for anything else is off-label.
What is the best peptide for diabetes?
There is no single answer. Our diabetes overview compares each candidate by evidence grade and legal status rather than by popularity.
Is orforglipron approved for type 2 diabetes?
The Foundayo label cited here approves it for weight management; the diabetes results come from trials.[10]
