Can retatrutide cause cancer?

No source we hold shows retatrutide causing cancer in people; the only retatrutide-specific cancer data are from mouse models, where weight loss on retatrutide reduced pancreatic tumour growth.

GLP-1 / GIP / glucagon triple agonistUpdated

Short answer

No source we hold shows retatrutide causing cancer in people; the only retatrutide-specific cancer data are from mouse models, where weight loss on retatrutide reduced pancreatic tumour growth.[1]

What the mouse studies found

In the pancreatic cancer mouse model, tumour volume fell 14-fold on retatrutide against 4-fold on semaglutide, in animals, not people.[1]

In a lung cancer mouse model, retatrutide halved tumour engraftment and cut tumour volume 17-fold compared with controls.[1]

The mice regained weight after retatrutide was withdrawn, yet the anti-tumour effect persisted, according to the authors.[1]

What human data exist

A meta-analysis of randomised trials of GLP-1 receptor agonists as a class, with retatrutide among the drugs, found no significant overall association with pancreatic cancer (relative risk 1.30, interval 0.86 to 1.97).[2]

The same analysis saw a higher pancreatic cancer risk when background medications were present (1.85) but not without them (0.81); it pools the whole drug class, not retatrutide alone.[2]

A review of unregulated injectables says investigational triple agonists such as retatrutide lack established human safety profiles and lists carcinogenesis among the potential risks of unregulated use.[3]
