The receptor semaglutide acts on
The Ozempic label describes semaglutide as binding to and activating the GLP-1 receptor, the target of the body's own GLP-1.[1]
The Wegovy tablets text puts the likeness to the natural hormone at 94% sequence homology.[2]
Blood-sugar lowering runs through insulin and glucagon, and both effects depend on glucose level.[1]
What the label says happens in the body
The Wegovy label links weight effects to appetite regions of the brain that carry GLP-1 receptors.[2]
Gastric emptying slows early after meals, which the Ozempic label says reduces how fast glucose enters the blood.[1]
In people with type 2 diabetes, semaglutide 1 mg cut fasting glucose by 29 mg/dL relative to placebo in the label's pharmacodynamic data.[1]
The Ozempic label reports the fasting glucagon fall at 8% and the 24-hour glucagon fall at 12% relative to placebo.[1]
Why the injection is weekly
Binding to albumin is the label's stated reason semaglutide lasts so long, because it lowers kidney clearance and protects against metabolic breakdown.[1]
The Ozempic label puts the elimination half-life at about 1 week and says the drug stays in circulation about 5 weeks after the last dose.[1]
The Wegovy label extends that to about 5 to 7 weeks after the last 2.4 mg or 7.2 mg injection or 25 mg tablet.[2]
What the label says is not fully understood
For liver disease (MASH), the Wegovy label says the precise mechanism is not fully understood and may involve several pathways mediated by weight loss and other factors.[2]

Common questions
How long does Semaglutide act in the body?
The half-life values we found are approximately 1 week (subcutaneous); approximately 1 week (subcutaneous and oral). A half-life is how long the amount in the body takes to fall by half; it is not the same as how long an effect lasts.
How does Ozempic work?
Ozempic is semaglutide; the label says it works by activating the GLP-1 receptor, which raises glucose-dependent insulin release and lowers glucagon.[1]
How does Wegovy work for weight?
Wegovy's label says the weight effect is likely mediated by affecting appetite.[2]
