What the labels say semaglutide is for
The Ozempic label lists type 2 diabetes blood-sugar control as the injection's indication, as an adjunct to diet and exercise.[27]
The Wegovy label ties its cardiovascular indication to adults who already have heart or vascular disease together with obesity or overweight, alongside diet and activity changes.[28]
The dated PubMed count for semaglutide and weight terms is 1978 human-only records and 160 animal records (2026-10-02); a count of papers is not a measure of benefit.
Calculated from goals file our PubMed count ledger; goal weight-loss; peptide semaglutide; query (semaglutide[tiab] OR Ozempic[tiab] OR Wegovy[tiab] OR Rybelsus[tiab] OR NN9535[tiab]) AND ("weight loss"[tiab] OR "weight reduction"[tiab] OR "body weight"[tiab] OR obes*[tiab] OR overweight[tiab] OR adiposity[tiab] OR "fat mass"[tiab] OR "body fat"[tiab] OR "fat loss"[tiab]); date 2026-10-02; human only 1978; animal 160. Formula: count of PubMed records for the dated query with the stated filter (humans not animals, and animals not humans)
Weight loss: what the trials measured
In a 68-week trial of adults with overweight or obesity, body weight fell 14.9% on semaglutide 2.4 mg against 2.4% on placebo.[12]
After a 20-week run-in, adults who kept taking semaglutide in the STEP 4 trial lost a further 7.9% over weeks 20 to 68, while those switched to placebo regained 6.9%.[13]
The cost side of that result: in the same trial, 4.5% of semaglutide participants stopped treatment for gastrointestinal events, against 0.8% on placebo.[12]
Heart and kidney claims, trial by trial
In SELECT, which enrolled adults with heart disease and overweight but no diabetes, a major cardiovascular event occurred in 6.5% on semaglutide and 8.0% on placebo.[9]
In adults with type 2 diabetes at high cardiovascular risk, the primary outcome occurred in 6.6% on semaglutide and 8.9% on placebo.[29]
In people with type 2 diabetes and chronic kidney disease, the risk of a major kidney-disease event was 24% lower on semaglutide than on placebo.[7]
That type 2 diabetes trial also reported more retinopathy complications on semaglutide, a harm that sits beside the heart result.[29]
Claims this page cannot confirm
A PubMed search dated 2026-10-02 for semaglutide with depression, suicide, psychiatric, Alzheimer or cognition terms in the title returned 159 records; this page cites none of them for a mood or memory benefit, so it gives no verdict on those claims.
Calculated from evidence file web/src/data/evidence/semaglutide.json; query semaglutide[tiab] AND (depress*[ti] OR suicid*[ti] OR "psychiatric"[ti] OR Alzheimer*[ti] OR cognit*[ti]); date 2026-10-02; count 159. Formula: count of PubMed records returned by the dated query
A PubMed search dated 2026-10-02 for semaglutide with inflammation, rheumatoid, psoriatic or C-reactive terms returned 60 records; none is quoted here, so no inflammation verdict is given.
Calculated from evidence file web/src/data/evidence/semaglutide.json; query semaglutide[tiab] AND (rheumatoid[tiab] OR "psoriatic"[tiab] OR "inflammation"[ti] OR "C-reactive"[ti]); date 2026-10-02; count 60. Formula: count of PubMed records returned by the dated query
One Alzheimer's programme did report a negative result: both phase 3 trials of oral semaglutide in early Alzheimer's disease were discontinued due to negative clinical outcome.[1]

Common questions
Is Semaglutide proven to work?
The strongest study we summarise for Semaglutide is human rct evidence, among 27 studies. A trial result is not an approval; see the legal status.
What does semaglutide do?
In the Ozempic label's own words, semaglutide lowers fasting and after-meal blood glucose and reduces body weight.[27]
Does semaglutide protect the heart?
The strongest heart evidence quoted here is a randomised trial in adults with established cardiovascular disease and overweight, where semaglutide 2.4 mg was superior to placebo for a composite of cardiovascular death, heart attack or stroke at a mean follow-up of 39.8 months.[9]
