Receptor and hormone cascade
Kisspeptin acts as the endogenous ligand of the G protein-coupled receptor 54 and regulates the hypothalamic-pituitary-gonadal axis.[3]
In men, kisspeptin-10 raised LH pulse frequency and secretory burst mass during infusion.[4]
In patients with a loss of neurokinin B signalling, continuous kisspeptin infusion restored pulsatile LH secretion.[5]
Why repeated dosing changes the response
In women with hypothalamic amenorrhea, repeated twice-daily injections caused the LH and FSH responses to fall.[6]
Twice-weekly dosing produced only partial desensitization compared with complete tolerance under twice-daily dosing.[7]
In men, the receptor stayed functional after 12 days of intermittent infusion, since a bolus still raised gonadotropins.[8]
Differences by sex and cycle phase
In women the response depends on cycle phase: the LH rise was largest before ovulation and smallest in the follicular phase.[3]
A comparison in healthy men and women found no gonadotropin change in the follicular phase at the maximal doses tested.[9]
Effects outside the reproductive axis
In mice, kisspeptin-10 caused tissue swelling and reduced skin blood flow, indicating vasoactive effects in the small vessels.[10]
In healthy women, kisspeptin-54 did not change growth hormone, prolactin or TSH over 7 days.[11]

Common questions
How long does Kisspeptin-10 act in the body?
The half-life values we found are mean elimination half-life 1.3-2.2 hours (subcutaneous (analog MVT-602)); median terminal elimination half-life 1.4-5.3 hours (subcutaneous (analog TAK-448)). A half-life is how long the amount in the body takes to fall by half; it is not the same as how long an effect lasts.
How does kisspeptin work?
It sits upstream of GnRH: kisspeptin stimulates GnRH neurons, and GnRH drives gonadotropin release.[4]
What is the mechanism of action of kisspeptin?
The receptor target is named, and the cascade runs from kisspeptin through GnRH to LH.[5]
